LAB SUMMARY:

Inoculate 5C sterile broth tube using aseptic technique

Complete the following steps:

1.Label sterile tubes with marker
2.Turn on Bunsen burner
3.Sterilize loop by placing into flame of Bunsen burner
4.Pick up bacterial culture tube. Remove cap. Briefly heat mouth of tube in Bunsen burner flame to reduce contamination
5.Insert loop into tube to acquire bacterial sample
6.Briefly heat mouth of tube, replace cap, and return tube to rack
7.Pick up sterile broth tube labelled 5C. Remove cap. Briefly heat mouth of tube in Bunsen burner flame to reduce contamination
8.Insert loop into tube
9.Briefly heat mouth of tube, replace cap, and place tube in appropriate incubation rack
10.Sterilize loop using the Bunsen burner. Return loop to holder

Inoculate remaining sterile broth tubes

Complete the following steps:

1.Select the 25C tube to automatically inoculate it with the bacterial culture
2.Select the 38C tube to automatically inoculate it with the bacterial culture
3.Select the 42C tube to automatically inoculate it with the bacterial culture
4.Select the 55C tube to automatically inoculate it with the bacterial culture
5.Select Incubate in order to incubate tubes for 24 hours at each temperature

Measure Growth with spectrophotometer

Complete the following steps for each sample tube listed:

Pick up pipette controller. Select pipette to attach to controller
Collect sample from tube and place into cuvette. Dispose of pipette in Sharps container
Place cuvette in spectrophotometer. Record measurement in Lab Data
Remove cuvette. Dispose of liquid in waste beaker and cuvette in Sharps container
